{"created":"2023-06-20T14:25:09.177307+00:00","id":216,"links":{},"metadata":{"_buckets":{"deposit":"11f55938-bbdc-409b-b3f1-eeb1e671be49"},"_deposit":{"created_by":3,"id":"216","owners":[3],"pid":{"revision_id":0,"type":"depid","value":"216"},"status":"published"},"_oai":{"id":"oai:dmu.repo.nii.ac.jp:00000216","sets":["81:29"]},"author_link":["752","751"],"item_2_biblio_info_12":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2004-03-25","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"1","bibliographicPageEnd":"44","bibliographicPageStart":"37","bibliographicVolumeNumber":"31","bibliographic_titles":[{"bibliographic_title":"Dokkyo journal of medical sciences"}]}]},"item_2_description_10":{"attribute_name":"抄録(日)","attribute_value_mlt":[{"subitem_description":"一般に,動物を用いた実験で,断頭後即座に取り出すことができない内耳のような組織の電子顕微鏡像を得るためには,全身灌流固定あるいは局所灌流固定のいずれかが必要である.しかし,これらの手法を用いると,血管内圧や外リンパ圧を人為的に上昇させることになり,血管内皮細胞や膜迷路の正しい形態を反映しない可能性がある.今回我々は,蝸牛にマイクロウエーブ照射固定(以下MWI固定と略す)法を用いた.モルモット断頭後蝸牛骨胞を切り出し,固定液(2%パラホルムアルデヒド, 0.5%グルタールアルデヒド混合液)に入れ,出力300Wで1分間照射を行い,照射後即座に4℃の固定液と交換した.この操作を10回繰り返し,計10分間照射した.固定液の液温は照射中30℃前後に保持し,照射後さらに1時間固定液中に浸した.固定液から取り出した蝸牛の外側壁の骨に僅かな裂隙を入れ,オスミウム酸で後固定した後,エポン樹脂に包埋した.標本は蝸牛軸を含む面で半切し,実体顕微鏡下にて蝸牛外側壁の骨を削除した後,超薄切片を作製して透過型電子顕微鏡で観察を行なった.血管条,コルヂ器の固定状態は従来の固定法と同等かそれ以上であった. MWIは手技が簡便であるばかりでなく,灌流固定の弱点を補う,すぐれた固定法であった.特に,硬組織に囲まれた軟組織の固定法として有望な方法である.","subitem_description_type":"Other"}]},"item_2_description_11":{"attribute_name":"抄録(英)","attribute_value_mlt":[{"subitem_description":"In animal experiment, total or local irrigation fixation must be generally used for the electronmicroscopic observation of the inner ear because the inner ear cannot be taken out just after decapitation. When these methods are used, the ultrafine structure of endothelial cells and membranous labyrinth may not be correctly reflected due to artificial increase of blood pressure or perilymph one. Now we conducted microwave irradiation (MWI) fixation of the inner ear in order to avoid these artifacts. The tympanic bullae of ten guinea pigs were fixed with MWI and those of five guinea pigs were fixed without MWI. The ultrafine structures of the organelles were kept with MWI, but in the control inner ear (without MWI) the cell membranes collapsed and organelles were severly damaged. And the MWI fixation was methodologically easier and faster than the irrigation fixation.
